Kfug:Have you purified your PPD preparation to see if the M , loo00 protein is the effective one? Lachmann: Purification is not easy, but if we couple PPD to polyaminostyrene, this picks up a small percentage of protein which is diazotizable and works well as a carrier. Williams: PPD contains two tyrosines. Have you attempted to find out whether you have labelled both, or one, and could you separate the three differently modified proteins possibly present? e. both residues are equivalent), whether they are inequivalent, or whether both are necessary.

PPD elicits delayed-type hypersensitivity (DTH) reactions in animals and humans who have previously encountered the tubercle bacillus, either as a result of infection or as a result of immunization with Bacillus Calmette-Guerin (BCG). Skin reactivity to PPD is widely used in this way as a diagnostic in uivo test t o detect previous exposure to tubercle bacilli. Even the repeated intradermal injection of tuberculin in a Mantoux test does not itself give rise to tuberculin sensitivity, so the test can be used on multiple occasions.

There must be non-specific processes involved. Ada: Does the increased traffic of lymphocytes cause tissue destruction in the lymph nodes, with the extra production of lymphokines by T cells? McConnell: You don’t see tissue destruction in the nodes, but there is extensive cell proliferation in the T cell-dependent areas. Corradin: The B cell has to process antigens, for the simple reason that T cells recognize peptides and B cells recognize native structures. We also know that for antibodies to be produced in vitro, a thousand times less antigen is needed than for T cell proliferation.
